Tokiomi Tohsaka, the guy with the goatee, is the current head of the Tohsaka clan. Kiritsugu's servant is Saber, a knightly girl with blonde hair. He and his wife wish to use the Grail to bring world peace. His wife is a member of the Einzbern family. Kiritsugu Emiya, the dark-haired guy that always smokes and has a hot albino wife, represents one of these clans, the Einzbern family. Of the 7 mages, three represent prestigious mage clans that date back to ancient time. The grail War consists of 7 mages, and 7 servants. You'll learn more about the specifics later.

The master of Caster is named Ryuunosuke, a serial killer with no magical talent who summoned his servant through the use of forbidden magical book. His servant is Iskandar, King of Conquerors. The master of Rider, Waver Velvet, is a motivated student seeking recognition for his own theories of magic by winning the war. He is in the war for his own personal gain. The master of Lancer, Kayneth Archibald El-Melloi, is a narcissistic and accomplished English mage. The crest worms will kill him very soon, so he seeks to win the war before then. Kariya has magical crest worms implanted in his body to allow him to use magic at a level to be recognized by the Grail (the Matou family excluding Zouken no longer has the ability to use powerful magic, hence why they adopted Sakura).
The father, Matou Zouken, seeks the Grail solely to acquire true immortality for his decaying body. He entered into the war at great personal injury to save Matou Sakura, the adoptive daughter of the family who was initially born to the Tohsaka family, from a fate as his father's pawn in the Fifth Grail War. The son of the third family, Matou Kariya, is the master of Berserker. Kirei is the master of Hassan, the Servant in the Assassin class, while Tohsaka is master of Archer (his true name is eventually revealed if it hasn't yet). Due to Kirei being a master, and connected to this plot, he is used as their accomplice in this goal. Tohsaka's goal is much more benign than the other masters, and the Church has decided to violate their neutrality and support him to make sure that the Grail is not won by a more greedy party. Tohsaka Tokiomi, the head of the second of the three families, has conspired with the Grail War's "referee" Kotomine Risei, and other master Kotomine Kirei (the son of Risei), to win the war through pooling their efforts. He is the "main character" of the story, so to speak. He has summoned King Arthur as his Servant, in the Saber class. To that end, he is willing to commit any atrocity to stop those in his way.
Emiya Kiritsugu, a magical assassin, works with the Einzbern family (one of the three families who originally created the Grail) in order to win the Grail War and wish for an end to human suffering and conflict.
